User jdhardy has proposed the issue:

When called on an existing directory, os.access (..., os.F_OK) succeeds on CPython but fails on IronPython. IronPython treats F_OK as "does this file exist", and then eventually calls File.Exists, which fails for directories.

It should be enough to change (in nt.access):

return context.LanguageContext.DomainManager.Platform.FileExists(path);


return context.LanguageContext.DomainManager.Platform.FileExists(path) || context.LanguageContext.DomainManager.Platform.DirectoryExists(path);"
