How about a few more clues -- are you running Apache or IIS?

If you are running Apache, on the first PC you may have entered this
into httpd.conf:
  ScriptAlias /mywiki/     "C:/......." 
when you should have entered:
  ScriptAlias /mywiki     "C:/......." 
e.g. no trailing slash.  The trailing slash is needed on the Alias
statements.
