Just my 2 cents:
I'm using wxPython and I'm impressed by the support that you can receive
from the community.
Robin Dunn, its author; usually replies to messages on the mailing list
promptly.
There is also the wxPython wiki that has a lot of informations and I've
found it very useful.
I also had no problem (on win32) in using it on different version of windows
(I could make an app of mine run on Win95 something I was really afraid of)
and in packaging a single exe file.

Another thought:
today I stumbled on fxPy a python wrapper for the Fox toolkit. The examples
really look good and I really love some of the not standard controls (a kind
of wheel) they use. It run with no problems on a standard Active Python
distro on win2k. But i never used it for serious development.

And finally:
with wxPython you can also give a try to PythonCard and boa contructor, both
are a kind of IDE even if very different in spirit and look.
