Not much differently from what you have already written :-) 

user_string= raw_input("Tell me:")
if user_string in 'y', 'ye', 'yes', 'yep':
	print "We are beginning"
elif user_string in 'n', 'no', 'nop', 'nope':
	print "You canceled"
