pyparsing <http://pyparsing.wikispaces.com/> is quite a cool package
for doing this sort of thing. Using your example:

#untested
from pyparsing import *

splitat = Or(":=", "+")
lexeme = Word(alphas)
grammar = splitat | lexeme

grammar.parseString("a:=b+c")
#returns (the equivalent of)  ['a', ':=', 'b', '+', 'c'].
