\ is used as a escape character in strings.
Use either \\ or a raw string, that is:

os.system('"C:\\Program Files\\GnuWin32\\bin\\tiff2pdf.exe" -o  
C:\\test.pdf C:\\test.TIF')
os.system(r'"C:\Program Files\GnuWin32\bin\tiff2pdf.exe" -o C:\test.pdf  
C:\test.TIF')
