I'm not Windows expert, but from my limited use (building the windows 
version of a cross-platform package) you don't normally specify the 
python at all. To build my software I do this:

cd <path to package>
setup.py py2exe

I have never found a way to specify the python, though there presumably 
is some way to do it.
